Alexandrium ostenfeldii growth and spirolide production in batch culture and photobioreactor ArchiMer

Growth and spirolide production of the toxic dinoflagellate Alexandrium ostenfeldii (Danish strain CCMP1773) were studied in batch culture and a photobioreactor (continuous cultures). First, batch cultures were grown in 450 mL flasks without aeration and under varying conditions of temperature (16 and 22 degrees C) and culture medium (L1, f/2 and L1 with addition of soil extract). Second, cultures were grown at 16 degrees C in 8 L aerated flat-bottomed vessels using L1 with soil extract as culture medium. Finally, continuous cultures in a photobioreactor were conducted at 18 degrees C in L1 with soil extract; pH was maintained at 8.5 and continuous stirring was applied. Batch ethanol fermentations from sweet sorghum juice by Saccharomyces cerevisiae NP 01 were carried out in a 500 ml air-locked Erlenmeyer flask under very high gravity (VHG) and static conditions. The maximum ethanol production efficiency was obtained when 9 g l-1 of yeast extract was supplemented to the juice. The ethanol concentration (P), productivity (Qp) and yield (Yp/s) were 120.24 ± 1.35 g l-1, 3.01 ± 0.08 g l-1 h-1 and 0.49 ± 0.01, respectively. Scale up ethanol fermentation in a 5-litre bioreactor at an agitation rate of 100 rev min-1 revealed that P, Qp and Yp/s were 139.51 ± 0.11 g l-1, 3.49 ± 0.00 g l-1 h-1 and 0.49 ± 0.01, respectively, whereas lower P (119.53 ± 0.20 g l-1) and Qp (2.13 ± 0.01 g l-1 h-1) were obtained in a 50-litre bioreactor....

Semi-scale production of PHAs from waste frying oil by Pseudomonas fluorescens S48 BJM

The present study aimed at developing a strategy to improve the volumetric production of PHAs by Pseudomonas fluorescens S48 using waste frying oil (WFO) as the sole carbon source. For this purpose, several cultivations were set up to steadily improve nutrients supply to attain high cell density and high biopolymer productivity. The production of PHAs was examined in a 14 L bioreactor as one-stage batch, two-stage batch, and high-cell-density fed-batch cultures. The highest value of polymer content in one-stage bioreactor was obtained after 60 h (33.7%). Whereas, the two-stage batch culture increased the polymer content to 50.1% after 54 h. Dentre os métodos disponíveis para avaliar o comportamento de agrotóxicos no solo, destacam-se o método "batch" (análise em batelada) e o deslocamento miscível. O objetivo deste estudo foi avaliar a sorção e a mobilidade do imazaquin em solos com diferentes características químicas, físicas e mineralógicas, utilizando as técnicas de deslocamento miscível e "batch". Os ensaios foram realizados no laboratório de Ecotoxicologia do Centro de Energia Nuclear na Agricultura da Universidade de São Paulo (CENA/USP), Piracicaba, São Paulo.
